Determination of fatty acids in fish oil
Abstract: This article analyzes and studies the contents of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in commercial fish oil capsule by combining with gas chromatography and mass spectrometry. The experiment of catalyst concentration, reaction temperature and time are optimized. Due to different derivation methods, the yield of EPA and DHA vary significantly. It gives the optimum ester exchange reaction conditions were as follows: with Sodium methoxide 1.0mol/L as catalyst, temperature 50 ℃, time of 20 minutes. The appropriate GC-MS oven program:keep at 50℃ for 5min, then 50℃/min to 150℃, and 10℃/min to 340℃, stay for 5min. With linoleic acid methyl ester as an external standard for quantitative analysis, we get the content for EPA and DHA (16.8% and 10.5% ), and the sum is between 20%-30%, which is high-quality fish oil. Therefore, this work introduces ways to discern the false fish oil capsule from the genuine by characteristic index of EPA and DHA in fish oil.
