摘要:杂环螺环化合物具有螺共轭、螺超共轭或异头效应等特殊性质,这些特殊性质使螺环化合物在医药、农药和材料等方面有着广泛的应用。
本文以二溴新戊二醇为原料来合成7-苯基-6,8-二氧杂螺[3.5]壬烷-2-羧酸。二溴新戊二醇经过在酸性条件下发生缩醛反应生成二氧杂环己烷,然后用NaH和丙二酸二乙酯在DMF溶剂反应,生成二酯化合物。在碱性的乙醇溶液中加热分解得到二酸化合物。最后在吡啶溶液中加热反应,得到7-苯基-6,8-二氧杂螺[3.5]壬烷-2-羧酸。含羧基的杂螺环化合物有更强的水溶性,作为药物成分,能够更好被人体吸收。而且螺[3.5]壬烷构架较稳定,可以作为有些不稳定化合物的替代物。该化合物的稳定性强和水溶性好的特点,也是作为抗菌剂的不可缺少特点。通过IR、1H NMR、13C NMR以及MS对合成的化合物进行了结构表征。22554 毕业论文关键词: 杂螺环;7-苯基-6,8-二氧杂螺[3.5]壬烷-2-羧酸;缩醛反应;合成
Synthesis of 7-phenyl-6,8-dioxaspiro[3.5] nonane-2- carboxylic acid
Abstract:Spiro heterocyclic compounds have spiroconjugation, snails hyperconjugation or extraordinary nature anomeric effect, these special properties make spiro-compounds have a wide range of applications in pharmaceuticals, pesticides and materials.
In this article, using 2,2-bis-(bromomethyl)-propane-1,3-diol as a starting material to synthesis 7 - phenyl -6,8 - dioxaspiro [3.5] nonane-2 - carboxylic acid. 2,2-bis-(bromomethyl)-propane-1,3-diol through acetalization under acidic conditions to produce dimethyl oxa-cyclohexane, and then was reacted with NaH in DMF and produces the diester compound. The diester compound was heated in an alkaline solution to generate diacid compound. Finally, the solution was heated in pyridine, to give 7-phenyl-6,8-dioxa-spiro[3.5]nonane-2-carboxylic acid. Spiro heterocyclic ring compound containing a carboxyl group are more soluble in water, as a pharmaceutical intermediate, which can be better absorbed by the body. And spiro [3.5] nonane framework structure is stable,and can be used as a substitute for some unstable compounds. The good stability and water-soluble of the compound also are indispensable features of antimicrobial agents. The compounds of structures are confirmed by IR, 1H NMR, 13C NMR and MS .
