摘要甘薯在生产上采用茎蔓扦插进行无性繁殖。扦插初期不定根的形成对甘薯苗后期存活和发育至关重要,其发生过程较为复杂并受一系列的信号分子调控。硝酸还原酶是介导一氧化氮(NO)信号产生的关键酶,其是否参与甘薯不定根形成仍不清楚。本实验主要目的为初步探究硝酸还原酶及NO信号在甘薯不定根发生时的作用。通过使用硝酸还原酶抑制剂(钨酸钠)、NO供体(硝普钠)和NO清除剂(cPTIO)处理甘薯茎蔓,实验发现硝酸还原酶抑制剂处理后甘薯不定根数量和长度均受到一定程度的抑制,说明硝酸还原酶在调控甘薯不定根发生中起到重要作用。此外,NO供体硝普钠处理对甘薯不定根的发生和不定根伸长也具有一定调控作用。当使用NO清除剂处理后发现,甘薯不定根数量和长度均显著被抑制。以上结果说明硝酸还原酶和NO信号参与了甘薯不定根的发生和不定根的伸长。本实验为今后进一步研究甘薯不定根发育机理奠定了一定的理论基础。77638

Study on the roles of nitrate reductase and nitric oxide in adventitious root formation and elongation in sweet potato

Abstract In the production of sweet potato, stem cuttings were used for asexual propagation。 The formation of adventitious root was very important to the survival and development of sweet potato in the later stage, and the process was complicated and regulated by a series of signal molecules。 Nitrate reductase is a key enzyme in the production of nitric oxide (NO) signal, and it is still not clear whether it is involved in the formation of the adventitious root of the sweet potato。 The purpose of this study was to explore the role of nitrate reductase and NO signal in the adventitious root of sweet potato。 By using of nitrate reductase inhibitor (tungstate), NO donor (sodium nitroprusside) and the NO scavenger c-PTIO, we explore the role of NR and NO in the formation of sweet potato adventitious roots。 The results showed that the nitrate reductase during adventitious root formation plays an important role。 At the same time, NO signal molecular also plays a role in this process。 The NO scavenger c-PTIO significantly inhibited adventitious root formation and elongation in sweet potato。 Our results proved that NR and NO signal play important roles in sweet potato adventitious root formation and elongation。 Our results supply some information for the study on the mechanisms of sweet potato adventitious root development。
