摘要:逆向脂肪酸β-氧化途径是合成中链脂肪酸最有效的途径。作为此途径的必须前体乙酰辅酶A却缺乏高效的合成途径。目前,合成乙酰辅酶A 的途径主要以敲除基因为主,但此途径不够高效。为此研究如何高效合成乙酰辅酶A并将其应用于中链脂肪酸的合成具有重要的意义。本文将讨论以乙酸盐转化成为乙酰辅酶A的方法来高效合成乙酰辅酶A。即探讨如何通过构建乙酰辅酶A前体途径,高效的表达乙酰辅酶A 合成酶,生产乙酰辅酶A,在提高胞内乙酰辅酶A含量的同时降低有毒副产物乙酸的积累,不影响菌体生长,从而提高中链脂肪酸的产量。29077 毕业论文关键词:乙酰辅酶A;乙酰辅酶A合成酶;中链脂肪酸
Effects of Acetyl-CoA synthetase in Synthesis of Medium Chain Fatty Acids
Abstract:The reversed fatty acid β-oxidation pathway is the most effective way to synthesize medium chain fatty acids. As a necessary precursor for this pathway: acetyl-CoA lacks an efficient synthetic pathway. At present, Gene Knockout is the main pathway to the synthesis of acetyl-CoA which is not efficient enough. It is of great significance to study how to synthesize acetyl-CoA efficiently and apply it to the synthesis of medium chain fatty acids. This article will discuss the conversion of acetate into acetyl-CoA to efficiently synthesize acetyl-CoA. Another way to say, we will increase the content of acetyl-CoA in the cells and reduce the accumulation of toxic byproduct acetic acid in the same time. Thereby, without affecting the growth of the cells, the yield of medium chain fatty acids is increased.
