摘要:红豆杉是我国的珍惜植物,研究发现,红豆杉中含有一种强效的抗癌物质——紫杉醇,以及一些紫杉烷类二萜化合物,如10-脱乙酰基巴卡丁III。这些紫杉烷类二萜化合物可以作为合成紫杉醇的原料。也就是说,如果我们可以合成紫杉醇的前体10-脱乙酰基巴卡丁III,那么紫杉醇的产量就可以大大提升。本次的论文采用了一些原本被视作废料的物质来合成目标产物,它们分别是7-木糖基-10-去乙酰基三尖杉宁碱、7-木糖醇-10-去乙酰基紫杉醇以及7-木糖醇-10-去乙酰基紫杉醇C。上述的三种物质作为原料,实验步骤依次为氧化水解、肼解、重结晶和精制。氧化水解的目的是脱去C-7位上的木糖基;肼解反应是为了使C-13位上的侧链被脱去。10-DABIII是合成紫杉醇的原料。本课题对提高紫杉醇产量、保护自然环境有着重大意义。22326 毕业论文关键词:紫杉醇;10-脱乙酰基巴卡丁III;木糖基;资源利用
The Synthesis Of 10-DeacetylbaccatinIII
Abstract: 10-DeacetylbaccatinIII,the precursor of the anticancer drug taxol and its analogue docetaxel, was obtained by treatment of a mixture of naturally occurring taxane glycosides in a relatively impurified form. The glycoside group of the taxane glycosides was deglycosidated to hydroxyl using periodate and followed hydrazine hydrate. The mixture was then hydrolyzed by hydrazine hydrate and purified through recrystallization. Since the taxane glycosides can be isolated during the process of the production of taxol without influencing on the productivity of the taxol itself, their conversion to 10-DABIII improves the total yield of taxol.
