摘要:脑膜炎致病菌穿透血脑屏障过程是病菌致脑膜炎的重要环节,脑微血管内皮细胞(Brain Microvascular Endothelial Cells,BMEC)是血脑屏障的重要组成部分,脑膜炎致病菌穿透血脑屏障往往伴随细菌侵袭脑微血管内皮细胞,因此本研究采用细胞侵袭实验初步筛选猪链球菌穿透血脑屏障相关基因。为了发掘SS2侵袭脑微血管内皮细胞能相关基因,本研究利用携带转座子TnYLB-1的pMar4s载体对ZY05719菌株基因组进行随机诱变,成功构建含100株的小型突变体文库,通过人脑微血管内皮细胞(hBMEC)侵袭实验筛选出1株侵袭能力减弱的突变株(P < 0.05)。实验结果表明,突变株的侵袭率与野生株相比降低了22.83倍,之后利用反向PCR技术对突变体中转座子的插入位点进行了鉴定,发现转座子插入位点位于细菌ABC转运通透酶基因(ZY05719_RS06545)内。本研究为进一步阐明SS2侵袭脑微血管内皮细胞机制奠定了基础。38796 毕业论文关键词:猪链球菌2型;毒力因子;侵袭;人脑微血管内皮细胞
Screening of virulence factors associated with invasion of brain micrvascular endothelial cells in Streptococcus Suis Serotype 2
Abstract: Meningitis pathogens crossing the blood-brain barrier process is the important step related with meningitis caused by the bacteria, brain microvascular endothelial cells is the important component of blood-brain barrier, so we used cell invasion experiments to screen related genes of Streptococcus suis through blood-brain barrier initially. In order to explore the related genes of SS2 to invade brain microvascular endothelial cells, the research randomly mutated the genome of ZY05719 strain by pMar4s vector carrying transposon TnYLB-1, and the mutant library containing 100 mutants was successfully constructed. One mutant (P < 0.05) with reduced ability to invade was screened by hMEBC invasion test. The result showed that the invasion rate of the mutant was 22.83 times lower than the wild strain, and then the insertion site of the transposon in the mutant was identified and analyzed by reverse PCR . The silencing gene inserted by transposon was found to be a ABC transporter permease gene (ZY05719_RS06545). This study will provide a foundation to the research of invasion of brain microvascular endothelial cells mechanism by SS2.
