摘要目的:探讨曲克芦丁对阻燃剂2,2′,4,4′-四溴联苯醚(BDE-47)诱导的小鼠肝脏NAD+耗竭的保护作用。方法: 将24只小鼠随机分为4组:正常对照组、BDE-47组(150mg/kg/d)、BDE-47(150mg/kg/d)+troxerutin组(150mg/kg/d)和troxerutin(150mg/kg/d)组,建立BDE-47诱导的小鼠肝脏损伤模型, 同时以曲克芦丁(troxerutin)处理, 12周后处死。检测各组小鼠体重、肝脏ALT活力、肝脏氧化应激指标(ROS水平)、肝脏NAD代谢酶含量、NAD+含量、PARP1活力变化。结果:BDE-47可导致小鼠肝脏的血清谷丙转氨酶( ALT)的活力水平升高,曲克芦丁能够使该酶水平降低。BDE-47能使小鼠ROS水平显著升高,引发氧化应激反应,曲克芦丁可以抑制氧化应激的发生。BDE-47可降低NAMPT的含量,升高PARP1的含量和其活力水平,从而使NAD+含量降低,曲克芦丁能够明显地升高NAMPT的含量,降低PARP1的含量和其活力水平,抑制NAD+的耗竭,减轻BDE-47诱导的小鼠肝脏损伤的影响,使肝细胞恢复至正常的状态。结论:曲克芦丁能抑制BDE-47诱发的氧化应激的反应, 从而保护肝脏NAD+的耗竭,最终改善毒物BDE-47对肝脏的损伤作用。50704

Troxerutin Protects against BDE-47-induced NAD+ Depletion in Mouse Liver

Abstract

Objective: In this research, we studied the protective effects of troxerutin on NAD+ depletion induced by BDE-47 in mouse liver. Methods: 24 mice were pided into four groups: control group, BDE-47 (150mg / kg / d) group, BDE-47 (150mg / kg / d) +troxerutin group (150mg / kg / d) and troxerutin (150mg / kg / d) group, then the mouse model of liver injury induced by BDE-47 was established, meanwhile mice were treated with troxerutin. After 12 weeks of treatment, all mice were killed. The body weight, glutamic pyruvic transaminase (ALT) activity of serum, ROS content and levels of hepatic NAD+ metabolic enzymes, as well as NAD+ content were determined in each group. Results: BDE-47 led to elevated levels of serum ALT in mice. However, troxerutin could decrease the level of ALT in BDE-47-treated mouse. BDE-47 markedly increased the ROS level in mouse liver, causing a severe oxidative stress. But troxerutin could inhibit oxidative stress in the livers of BDE-47-treated mice. Furthermore, BDE-47 significantly reduced NAMPT level, increased protein level and activity of PARP1, thereby decreased NAD+ content in mouse liver. However, troxerutin notably improved the levels of these NAD+ metabolic enzymes and the content of NAD+.  Conclusion: Troxerutin could inhibit oxidative stress, thereby attenuated NAD+ depletion, ultimately protected mouse against BDE-47-induced liver injury.
