摘   要以构巢曲霉(Aspergillus niger)作为实验材料,以其新发现的sHSP7和HSP81这两个热激蛋白作为研究对象,分析它们在不同的发育水平、冷热压力、渗透压和氧化压力条件下基因表达水平的差异,从而弄清这2个sHSP的遗传进化起源差异。处理条件如下:71154

(1)不同渗透胁迫条件下,两种热激蛋白的基因表达水平以及它们之间的差异;

(2)不同发育水平条件下,两种热激蛋白的基因表达水平以及它们之间的差异;

(3)不同冷热压力条件下,两种热激蛋白的基因表达水平以及它们之间的差异;

(4)不同氧化压力条件下,两种热激蛋白的基因表达水平以及它们之间的差异;

(5)不同营养水平条件下,两种热激蛋白的基因表达水平以及它们之间的差异;

实验结果表明:除了在NaCl-15渗透压处理菌体时,sHSP7的基因表达上调与HSP81的基因表达刚好相反外,其他三种渗透胁迫下它们的基因表达都是相同的趋势,所以它们很可能是相同的进化;不同发育水平条件下,sHSP7和HSP81的基因表达都是相同趋势,所以它们很可能是相同的进化;在不同冷热压力下,sHSP7和HSP81的基因表达都是相同趋势,所以它们很可能是相同的进化;在不同氧化压力条件下,sHSP7和HSP81的基因表达都是相同趋势,所以它们很可能是相同的进化;在不同营养水平条件下,sHSP7和HSP81的基因表达都是相同趋势,所以它们很可能是相同的进化。

Abstract Use Aspergillus nidulans as experimental material.The two newly discovered Heat shock proteins sHSP7 and HSP81 were used As the research object.Analysis their differences in gene expression level at different developmental levels, cold and heat stress, osmotic pressure and oxidative stress conditions,so as to make clear the differences of the genetic evolution of the 2 HSP.Treatment conditions are as follows:

(1)Under different osmotic stress conditions, gene expression levels of two kinds of heat shock proteins and the differences between them.

(2)Under different developmental level conditions, gene expression levels of two kinds of heat shock proteins and the differences between them.

(3)Under different hot and cold pressure conditions,gene expression levels of two kinds of heat shock proteins and the differences between them.

(4)Under different oxidative stress conditions,gene expression levels of two kinds of heat shock proteins and the differences between them.

(5)Under different nutritional conditions,gene expression levels of two kinds of heat shock proteins and the differences between them.

    Experimental results show that:In addition to the NaCl-15 osmotic pressure in the treatment of bacterial,The gene expression of sHSP7 was up-regulated and the expression of HSP81 was just opposite.The expression of their genes was the same in the other three kinds of osmotic stress.So they are likely to be the same.Under different developmental level conditions,the gene expression of sHSP7 and HSP81 was the same trend.So they are likely to be the same.Under different hot and cold pressure conditions,the gene expression of sHSP7 and HSP81 was the same trend.So they are likely to be the same.Under different oxidative stress conditions,the gene expression of sHSP7 and HSP81 was the same trend.So they are likely to be the same.Under different nutritional conditions,the gene expression of sHSP7 and HSP81 was the same trend.So they are likely to be the same.
