摘要: 本课题论文以中国传统食品——金华火腿为研究对象,将预处理后的金华火腿通过多次煮沸、旋转蒸发后得到金华火腿水溶性粗提物,再将粗提物进行超滤,收集分子量在5000Da以下的分离物,最后将其分离物经由凝胶层析柱分离纯化,收集得到的四种分离物。采用DPPH、ABTS、羟自由基和还原力法四种不同的体外抗氧化方法对火腿的粗提物、分离后提取物进行抗氧化性质的测定。通过对比研究发现:经由超滤提取分离后的提取物的抗氧化性明显高于粗提物,同时采用ABTS和还原力法对相同蛋白含量条件下四种分离物组分的抗氧化能力的比较研究表明,第三种分离物组分的抗氧化能力相较于其它分离物更强。7356

Antioxidant capacity of Crude extract from ham
Abstract:Jinhua ham, Chinese traditional food, was selected as research material in this article. Firstly, water-soluble crude extract from Jinhua ham was obtained after several boiling and evaporations on primed Jinhua ham. Then, the above extract was dissolved into water and ultra-filtrated through a membrane to get separations with a molecule weight under 5000Da. The collected solution was added into chromatography column to collect four different matters existing in the four peaks, accordingly. Finally, four different methods, which were DPPH, ABTS, reducing power method and hydroxyl radical, were utilized to measure antioxidant ability of crude and secondary extract. After comparison of antioxidant capacity of crude extract and secondary extract, data obtained through experiments showed that antioxidant ability of secondary extract gotten through ultrafiltration was much higher than that of crude extract. At the same time, the two methods ABTS and reducing power were used to measure antioxidant strength of four separations with the same protein content. Results suggested that antioxidant capacity of the third separation was much stronger and more obvious than the others’.
