摘要:本试验旨在研究姜黄素、去甲氧基姜黄素和双去甲氧基姜黄素三种姜黄素类化合物对于水溶性自由基2,2-偶氮二(2-脒基丙烷)盐酸盐(AAPH)诱导的红细胞氧化损伤的保护作用。试验选取鸡红细胞为试验对象,设置0.5-10 μM一共5个浓度梯度,通过水溶性自由基AAPH诱导红细胞建立氧化应激模型,分别在37°C条件孵育下处理3h、4h、5h,测定红细胞溶血率、丙二醛(MDA)浓度。结果表明:(1)姜黄素类化合物能有效抑制AAPH诱导的红细胞溶血作用;其中,双去甲氧基姜黄素的效果最为显著。(2)姜黄素类化合物能有效降低红细胞中MDA浓度。其中姜黄素的效果最为明显,去甲氧基姜黄素次之,双去甲氧基姜黄素较其他两种姜黄素类化合物效果最差。综上所述,姜黄素类化合物具有抗氧化功能,对于红细胞的氧化损伤具有保护作用,三种姜黄素的抗氧化效果具有一定差异。25195 毕业论文关键词:姜黄素;去甲氧基姜黄素;双去甲氧基姜黄素;红细胞;氧化损伤
Protective effects of curcuminoids on oxidative damage in erythrocytes
Abstract:The aim of this study was to investigate the protective effects of curcuminoids on oxidative damage induced by AAPH in chicken erythrocytes at concentrations ranging from 0.5 μM to 10 μM. The hemolysis and MDA concentrations were determined at 3, 4, 5 h of incubation at 37°C. The results showed that curcuminoids prevented the AAPH-challenged erythrocyte membrane from hemolysis while bisdemethoxycurcumin showed a higher inhibitory power of hemolysis. Curcuminoids significantly decreased MDA concentrations induced by AAPH treatment. The positive effects of curcuminoids on MDA concentrations decreased in the following order: curcumin > demethoxycurcumin > bisdemethoxycurcumin. In conclusion, curcuminoids showed potential antioxidant activities with varying extent, and were effective in the protection of erythrocytes against oxidant damage.
