摘要:为快速检测疫苗产品中的外源猪圆环病毒2型(PCV2),针对其ORF2基因设计一对引物Y1/Y2进行PCR检测,并从其特异性、敏感性等方面与PCV国家标准引物(P1/P2/P3)和PCV出入境引物(nP1/nP2/nP3/nP4)进行比较。结果显示:三种引物均具有很高的特异性;用大北农株(DBN-SX07)验证三种引物的检测敏感度,引物Y1/ Y2最高检测敏感度为0.19TCID50,引物P1/ P2/ P3最高检测敏感度为0.079 TCID50,引物nP1/ nP2/ nP3/ nP4最高检测敏感度为7.9 TCID50;临床株验证时,引物Y1/ Y2最高检测敏感度为1.58×10-6TCID50,引物P1/ P2/ P3最高检测敏感度为1.58×10-4TCID50,,引物nP1/ nP2/ nP3/ nP4最高检测敏感度为1.58×10-4TCID50;在高稀释度时,出入境引物检验结果判定总是出现混乱,存在一定缺陷。结果表明,引物Y1/Y2检测PCV2敏感性更高,更适于本公司质检部门进行疫苗产品的外源PCV2检测。26601 毕业论文关键词:猪圆环病毒2型;不同引物;PCR;敏感性
Comparative study on the sensitivity of three kinds of different primers for the detection of Porcine circovirus type 2
Abstract:In order to detect exogenous porcine circovirus type 2 (PCV2) in vaccine products rapidly, we designed a pair of primers Y1 and Y2 on the basis of the ORF2 gene for PCR detection,and compared its specificity, sensitivity with PCV national standard primers (P1/P2/P3) and entry-exit inspection primer. The results showed that (1) Three kinds of primers had high specificity for the detection of PCV2; (2) When the DBN-SX07 strain was detected , the PCRs using primers Y1/Y2 and P1/P2/P3 were able to detect at least 0.079TCID50 PCV2, while the nest-PCR primers nP1/nP2/nP3/nP4 could detect only 7.9 TCID50 ; (3) When the clinically acquired strain was been detected, the PCR using primers Y1/Y2 was able to detect at least 1.58×10-6TCID50 PCV2, while the primers P1/P2/P3 and nP1/nP2/nP3/nP4 could detect only 1.58×10-4TCID50. (4) When the virous titer is high, the results of the nest-PCR primers nP1/nP2/nP3/nP4 are always confused. Consequently, the primers Y1/Y2 had higher sensitivity for the detection of PCV2, and it was more suitable for the detection of exogenous PCV2 in vaccine products.
