摘 要:氧化石墨烯(GO)具有良好的水溶性和生物相容性,并且可以吸附DNA单链并猝灭链上荧光染料的荧光,从而在生物分子的检测中应用广泛。本实验中,我们设计了一条单链DNA探针,探针的5'端荧光标记,另一端序列可以自杂交形成小发夹结构,从而形成了一个发夹引物。在目标聚合酶和dNTPs混合物存在下,发生聚合延伸形成双链DNA产物,加入GO后,仍然得到较强的荧光信号。没有聚合酶不能发生聚合延伸反应,荧光发夹引物会被GO吸附猝灭,从而实现对目标聚合酶KF活性的检测。该方法简便快速、灵敏高,还可以进一步用作聚合酶抑制剂的筛选。9231

Fluorescent Hairpin Primer for Assay of DNA polymerase Based on Graphene Oxide and Polymerization Extension
Abstract: Graphene oxide (GO) has an excellent solubility and biocompatibility, which can also adsorb the single-stranded DNA (ssDNA) and quench its fluorescence, thus has found a wide application in biomolecular detection. In this paper, we synthesized a 5' dye-labeled ssDNA probe, and it can self-hybridize into a hairpin primer (HP). In the presence of target polymerase and dNTPs, HP can undergo polymerase elongation and generate double-stranded DNA (dsDNA). Upon addition of GO, it can also obtain a strong fluorescence signal. While the polymerization extension won’t occur in the absence of target polymerase, and the HP will be adsorbed and quenched by GO. By monitoring the fluorescence change of this system, the detection of polymerase activity can be realized. The proposed method is simple, rapid and high sensitive, which can also be used for screening the inhibitors of polymerase.
