摘要:抗生素的发现对人类和动物感染性疾病发挥了重要作用。然而近年来抗生素在人医临床、农业,尤其是畜牧业的滥用,导致了耐药菌的大范围爆发,如今已成为了一个严峻的全球性问题。作为抗生素主要生产菌和抗生素耐药基因根源的土壤微生物中蕴含了大量未知的耐药基因,在一定条件下它们能够在不同环境介质和微生物之间迁移和传播,因而对临床微生物和人类健康构成了极大的潜在威胁。本课题应用功能筛选方法从宏基因组文库中筛选头孢菌素类抗生素耐药基因,成功构建阳性亚克隆,并选取头孢他啶阳性亚克隆MQ-329、MQ-937,对功能基因进行异源表达,并初步验证了其耐药功能。38802 毕业论文关键词:土壤宏基因组;β-内酰胺酶;异源表达;功能分析
Heterologous Expression and Functional Analysis of β - Lactamases from Soil Microorganisms
Abstract:The discovery of antibiotics has played an important role in human and animal infectious diseases. However, in recent years antibiotics in human medicine clinical, agricultural, especially animal husbandry abuse, leading to a wide range of pathogens in drug resistance phenotype outbreak, has now become a serious global problem. As a major producer of antibiotics and antibiotic resistance gene roots in the soil microbial contains a large number of unknown resistance genes, under certain conditions they can migrate and spread in different environmental media and microbial migration, thus posing a significant potential threat to clinical microbes and human health. In this study, the functional screening method was used to screen the cephalosporin antibiotic resistance gene from the macro genomic library, and successfully constructed positive subclones, and the cephalosporin-positive subclones MQ-329 and MQ-937 were selected to analyze the heterologous expression and function of the resistant gene, finally, analysis of the biological function of new drug resistance genes from soil microorganism from protein function.
