摘要:内生真菌作为特殊真菌类群,广泛存在于自然界中,几乎所有植物内都含有内生真菌。内生真菌通过代谢产生大量活性物质,例如杀虫、抗菌、抗肿瘤等活性物质,一部分已在临床上应用,效果显著。本课题的工作主要包括从南通、安徽等地采集的三十五种植物标本中,采用组织分离法分离出320种植物内生真菌。 用它们的代谢产物做活性检测后(即使用抗肿瘤体外细胞毒筛选模型MTT法),获得1株内生真菌对A549细胞的生长存在最强的抑制作用。通过对高抗肿瘤活性菌株SK11359043的DNA提取,扩增其ITS 测序,在数据库进行Blast,并结合菌丝形态和产孢结构鉴定目标内生真菌的分类地位。依据菌株的初步鉴定结果,判定菌株SK11359043为茎点霉属 Phoma sp. 38407 毕业论文关键字:植物内生真菌;抗肿瘤;菌种鉴定;
The separation and purification of endophytic fungi and the antitumor activity screening of their metabolic products
Abstract: Endophytic fungi are widely distributed in nature in almost all plants as a special category of Fungi. They can produce abundant metabolites including insecticidal, antimicrobial and antitumor active substances. Part has been applied in clinical for its significant effect . 320 species of plant endophytic fungi were isolated from 32 kinds of plant specimens picking from Nantong and Anhui . The Antitumor activity of their metabolic products to A549 tumor cells in vitro were screened using MTT method . The results showed that SK11359043 has the best significant inhibitory activities to A549 cells. DNA was extracted from strain SK11359043. Its ITS was amplified and sequenced in Shanghai Sangon Biotech (Shanghai).The sequence was blasted in the NCBI database. The classification was identificated combining with the morphology of hyphae and spores of endophytic fungi. The results showed that strain SK11359043 belong to Phoma sp.
